Cellobiose dehydrogenase (CDH) [E. C. 1.1.99.18] is an extracellular enzyme which is produced by various wood degrading fungi. The enzyme has several technical applications like in- biosensors, bioremediation or biocatalysis. I have explored the fungus Termitomyces clypeatus for CDH production. The enzyme has been purified and physicochemically characterized. It was used for the production of lactobionic acid- a constituent in the ‘Wisconsin transplantation used during organ preservation for transplantation. The availability of large amount of this enzyme is a prerequisite for successful biotechnological applications. Lastly, efforts for large-scale production of the enzyme have been made through fermentation technics with cost-effectiveness.

ONGOING RESEARCH PROJECT
One year research project-
“Isolation and purification of xanthine oxidase from bovine milk and screening of new
chemical entities (NCEs) as xanthine oxidase inhibitors”
Xanthine oxidase (metalloflavoprotein) is an enzyme (hyperactivity) involved in the gout and various free radical
mediated cancers so it can be a target for gout and different cancers. For this I have isolated and purify the xanthine
oxidase from bovine milk and screen out the different NCEs (provided by Mr. Srikant in dept. of medicinal chemistry
under Dr. A. K. Chakraborti) after setting different kinetic parameters of xanthine oxidase. The different methodologies
and proceduce adopted to perform whole project, are
	 Isolation of enzyme by ammonium sulfate precipitations, dialysis and by using centricon
	 Purification by the hydroxyappetite (adsorption) chromatography column and by SDS-PAGE
	 Enzyme activity and specific activity measurement by ELISA-plate reader and double beam
	spectrophotometer
	Kinetic study of enzyme done by double beam spectrophotometer
	Screening and inhibitory kinetic study of different NCEs is also done by double beam spectrophotometer

“Isolation and purification of xanthine oxidase from bovine milk and screening of new<br />
chemical entities (NCEs) as xanthine oxidase inhibitors”<br />
Xanthine oxidase (metalloflavoprotein) is an enzyme (hyperactivity) involved in the gout and various free radical<br />
mediated cancers so it can be a target for gout and different cancers. For this I have isolated and purify the xanthine<br />
oxidase from bovine milk and screen out the different NCEs (provided by Mr. Srikant in dept. of medicinal chemistry<br />
under Dr. A. K. Chakraborti) after setting different kinetic parameters of xanthine oxidase. The different methodologies<br />
and proceduce adopted to perform whole project, are<br />
	 Isolation of enzyme by ammonium sulfate precipitations, dialysis and by using centricon<br />
	 Purification by the hydroxyappetite (adsorption) chromatography column and by SDS-PAGE<br />
	 Enzyme activity and specific activity measurement by ELISA-plate reader and double beam<br />
	spectrophotometer<br />
	Kinetic study of enzyme done by double beam spectrophotometer<br />
	Screening and inhibitory kinetic study of different NCEs is also done by double beam spectrophotometer</p>

<p>Career Goal:<br />
I look myself as a researcher who wants to understand the microbial world and come up with solution which will be useful to humanity at large. My present work revolves around understanding the complex mechanism of microbial pathogenesis.
